Anyways, here's my first recommendation: Fullmetal Alchemist (FMA).
FMA is set in a world similar to our own, but instead of developing electronically, alchemy was advanced. The story follows two alchemist brothers, Edward and Alphonse, who are on a quest to find the Philosopher's Stone. When they were younger, their mother died (Dad disappeared long ago), so they tried to bring her back to life using alchemy. It backfired, horribly, ending up with Ed losing an arm and a leg, and Al his entire body. They think finding the Stone could bring back their intact bodies back, but it's not that simple. From there, you'll have to watch the anime!
As for genre, it's action/comedy/adventure/tragedy (just a little). It was based of the manga FMA, but the original anime takes a really dark turn away from the manga in the second season. There was a second anime made, FMA Brotherhood. This one follows the manga pretty closely, and doesn't depend on the original. There are a couple movies that go along with the anime. One, Conqueror of Shamballa, is a continuation of the original anime and completes the story, so watch it afterwards. It's pretty dark and has a really bittersweet ending. The other, The Sacred Star of Milos, falls around the 24th episode of FMA Brotherhood. Watch it after the end of the anime to prevent the collapse of your heart from lack of FMA.
